SOP for Mixing Tribromoethanol (TBE, Avertin®)
General information
- For use in mice only
- Can be used only ONCE in any individual animal
- Requires additional justification in an animal use protocol as it does not reflect current standard of veterinary care
- Contact your research animal veterinarian for what may be considered adequate justification for use
Materials needed:
- 2,2,2-tribromoethyl alcohol
- tertiary amyl-alcohol
To make a stock of 100% TBE:
- 10g of 2,2,2-tribromoethyl alcohol
- 10 ml tertiary amyl-alcohol
- Fully dissolve (heat to 50C, use a magnetic stirrer overnight)
- Store in a dark or foil-covered container at 4C
- DO NOT USE IF DISCOLORED OR PRECIPITATE IS PRESENT
- Discard stock after 4 months
- Label all containers with name of drug, concentration, expiration date (4 months after prep), initials of preparer
Working solution:
- Dilute 100% stock to 1.2-2.5% v/v in water or isotonic saline
- Stir vigorously until dissolved
- Filter sterilize through 0.2 micron filter
- Make fresh working solution immediately prior to experiment
- pH of final solution must be >5
- Label all containers with name of drug, concentration, date prepared, initials of preparer
- Dose is 125-250 mg/kg given intraperitoneally—for a 1.25% solution, this is 0.02 ml/g body weight
